Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000465.4(BARD1):c.163A>C (p.Asn55His), citing Ambry Variant Classification Scheme 2023: The p.N55H variant (also known as c.163A>C), located in coding exon 2 of the BARD1 gene, results from an A to C substitution at nucleotide position 163. The asparagine at codon 55 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
