This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 3995, where T is replaced by G; at the protein level this means replaces isoleucine at residue 1332 with serine — a missense variant. Submitter rationale: This missense variant replaces isoleucine with serine at codon 1332 of the ATM protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. In a large international case-control study, this variant was reported in 1/60465 breast cancer cases and 0/53461 controls (PMID: 33471991). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
