Submitter rationale: The c.264delC deletion in the PURA gene has not been reported previously as a pathogenic variantnor as a benign polymorphism, to our knowledge. The c.264delC deletion replaces thelast 235 amino acid residues in the PURA-encoded protein with 136 aberrant residues and is expected toresult in protein truncation and to alter normal protein structure and function. The c.264delC variant wasnot observed in approximately 6500 individuals of European and African American ancestry in the NHLBIExome Sequencing Project, indicating it is not a common benign variant in these populations.We interpret c.264delC as a pathogenic variant.
