Pathogenic — the classification assigned by GeneDx to NM_000444.6(PHEX):c.1719dup (p.Ile574fs), citing GeneDx Variant Classification (06012015). This variant lies in the PHEX gene (transcript NM_000444.6) at coding-DNA position 1719, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 574, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.1719dupT duplication in the PHEX gene causes a frameshift starting with codon Isoleucine 574,changes this amino acid to a Tyrosine residue and creates a premature Stop codon at position 8 of the newreading frame, denoted p.Ile574TyrfsX8. This suplidation is predicted to cause loss of normal proteinfunction either through protein truncation or nonsense-mediated mRNA decay. Although this variant hasnot been previously reported to our knowledge, we interpret it as pathogenic.
