NM_000368.5(TSC1):c.658G>C (p.Val220Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V220L variant (also known as c.658G>C), located in coding exon 5 of the TSC1 gene, results from a G to C substitution at nucleotide position 658. The valine at codon 220 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
