Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.631G>C (p.Glu211Gln), citing Ambry Variant Classification Scheme 2023: The p.E211Q variant (also known as c.631G>C), located in coding exon 5 of the TSC1 gene, results from a G to C substitution at nucleotide position 631. The glutamic acid at codon 211 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
