NM_017636.4(TRPM4):c.2476T>G (p.Cys826Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C826G variant (also known as c.2476T>G), located in coding exon 17 of the TRPM4 gene, results from a T to G substitution at nucleotide position 2476. The cysteine at codon 826 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
