This variant lies in the LAMB1 gene (transcript NM_002291.3) at coding-DNA position 2724, duplicating one base; at the protein level this means shifts the reading frame starting at glycine residue 909, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.2724dupT variant in the LAMB1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.2724dupT variant causes a frameshift starting with codon Glycine 909, changes this amino acid to a Tryptophan residue, and creates a premature Stop codon at position 39 of the new reading frame, denoted p.Gly909TrpfsX39. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.2724dupT variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.2724dupT as a likely pathogenic variant.
