Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001365276.2(TNXB):c.9031G>A (p.Glu3011Lys), citing Ambry Variant Classification Scheme 2023: The p.E3009K variant (also known as c.9025G>A), located in coding exon 25 of the TNXB gene, results from a G to A substitution at nucleotide position 9025. The glutamic acid at codon 3009 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
