NM_004655.4(AXIN2):c.512T>C (p.Phe171Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F171S variant (also known as c.512T>C), located in coding exon 1 of the AXIN2 gene, results from a T to C substitution at nucleotide position 512. The phenylalanine at codon 171 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
