This variant lies in the NOD2 gene (transcript NM_001370466.1) at the canonical splice donor site of the intron immediately after coding-DNA position 2465, duplicating one base. Submitter rationale: The NOD2 c.2546+2dup variant (rs748508924), to our knowledge, is not reported in the medical literature but is reported in ClinVar (Variation ID: 418716). This variant is observed in the general population with an overall allele frequency of 0.01% (36/282564 alleles) in the Genome Aggregation Database. Computational analyses (Alamut v.2.11) predict that this variant may impact splicing by disrupting the nearby canonical donor splice site. However, loss-of-function variants are not an established mechanism of disease for Blau syndrome, but have been associated with an increased risk for Crohn's disease (Huang 2017).
