Uncertain significance for RASopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005633.4(SOS1):c.3511-14_3511-10del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SOS1 gene (transcript NM_005633.4) at 14 bases into the intron immediately before coding-DNA position 3511 through 10 bases into the intron immediately before coding-DNA position 3511, deleting this region. Submitter rationale: This sequence change falls in intron 22 of the SOS1 gene. It does not directly change the encoded amino acid sequence of the SOS1 protein. This variant is present in population databases (rs760713912, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with SOS1-related conditions. ClinVar contains an entry for this variant (Variation ID: 418501). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
