NM_000314.8(PTEN):c.244A>G (p.Asn82Asp) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): This variant is denoted PTEN c.244A>G at the cDNA level, p.Asn82Asp (N82D) at the protein level, and results in the change of an Asparagine to an Aspartic Acid (AAT>GAT). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. PTEN Asn82Asp was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Asparagine and Aspartic Acid differ in some properties, this is considered a semi-conservative amino acid substitution. PTEN Asn82Asp occurs at a position that is highly conserved across species and is located within the Phosphatase tensin-type domain (UniProt, Nguyen 2013). In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, it is unclear whether PTEN Asn82Asp is pathogenic or benign. We consider it to be a variant of uncertain significance.
