This variant lies in the PTEN gene (transcript NM_000314.8) at 15 bases upstream of the translation start (5' untranslated region), where C is replaced by T. Submitter rationale: This variant causes a C to T nucleotide substitution at the -15 position of the 5'UTR of the PTEN gene. To our knowledge, RNA studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/251476 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
