NM_138395.4(MARS2):c.175C>T (p.His59Tyr) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): A novel H59Y missense change that is likely pathogenic was identified in the MARS2 gene. It has not been published as a pathogenic variant, nor has it been reported as a benign polymorphism to our knowledge. The amino acid change is non-conservative in that a positively charged Histidine residue is replaced by an uncharged Tyrosine residue. This change occurs at a highly conserved position in the MARS2 protein, and multiple in-silico analysis programs predict that H59Y is damaging to the MARS2 protein. Therefore, H59Y is a strong candidate for a pathogenic variant, however the possibility that it is a benign variant cannot be excluded.
