Likely pathogenic — the classification assigned by GeneDx to NM_000215.4(JAK3):c.218T>G (p.Leu73Arg), citing GeneDx Variant Classification (06012015): A novel L73R missense change likely associated with SCID was identified in the JAK3 gene. To our knowledge, this variant has neither been published as a pathogenic variant, nor reported as a benign polymorphism. L73R represents a non-conservative amino acid substitution as a non-polar Leucine residue is replaced with a large, positively-charged Arginine residue. This substitution occurs at a position in the JAK3 protein that is highly conserved among species. Therefore, L73R is a strong candidate for a pathogenic variant however, the possibility that it is a benign variant cannot be excluded.
