Pathogenic — the classification assigned by GeneDx to NM_000051.4(ATM):c.2167_2173del (p.Val723fs), citing GeneDx Variant Classification (06012015): This deletion of seven nucleotides in ATM is denoted c.2167_2173delGTGGGTG at the cDNA level and p.Val723SerfsX10 (V723SfsX10) at the protein level. The normal sequence, with the bases that are deleted in brackets, is TTTG[GTGGGTG]TCCT. The deletion causes a frameshift, which changes a Valine to a Serine at codon 723, and creates a premature stop codon at position 10 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. we consider this variant to be pathogenic.
