NM_000051.4(ATM):c.56G>A (p.Arg19Lys) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): This variant is denoted ATM c.56G>A at the cDNA level, p.Arg19Lys (R19K) at the protein level, and results in the change of an Arginine to a Lysine (AGA>AAA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Arg19Lys was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Arginine and Lysine share similar properties, this is considered a conservative amino acid substitution. ATM Arg19Lys occurs at a position that is moderately conserved across species where Arg to Lys substitutions are accepted in several vertebrate species. This variant is not located within a known functional domain and in silico analyses predict that it is unlikely to alter protein structure or function. Based on currently available information, it is unclear whether ATM Arg19Lys is pathogenic or benign. We consider it to be a variant of uncertain significance.
