NC_000016.10:g.(?_23621362)_(23641357_?)del was classified as Pathogenic for Familial cancer of breast by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-10 of the PALB2 gene, which includes the initiator codon. The 3' boundary is likely confined to the intronic region between exons 10 and 11. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. Truncating variants in PALB2 are known to be pathogenic. A similar deletion encompassing exons 1-10, in trans with a second PALB2 truncating variant, was reported in a patient affected with autosomal recessive Fanconi anemia (PMID: 17200672). For these reasons, this variant has been classified as Pathogenic.
