NC_000001.11:g.(?_17018722)_(17018958_?)del was classified as Uncertain significance for Gastrointestinal stromal tumor; Pheochromocytoma/paraganglioma syndrome 4; Pheochromocytoma by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 8 of the SDHB gene. The 5' boundary is likely confined to intron 7. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated SDHB protein. Deletions of exon 8 have not been reported in the literature in individuals with a SDHB-related disease. While this deletion is expected to result in a truncated SDHB protein, the effects of this on SDHB protein function are unknown. Therefore, it has been classified as a Variant of Uncertain Significance.
