Pathogenic for Supravalvar aortic stenosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000007.13:g.(?_73442119)_(73484237_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the ELN gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Gross deletions of the entire ELN gene have been reported in individuals affected with isolated supravalvular aortic stenosis, as well as in individuals affected with Williams-Beuren syndrome (PMID: 7693128, 7726172). Williams-Beuren syndrome is associated with the heterozygous deletion of 1.5 to 1.8Mb on chromosome 7q11.23, which includes the ELN gene. For these reasons, this variant has been classified as Pathogenic.
