Uncertain significance for Familial adenomatous polyposis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.10:g.(?_112828856)_(112828972_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 14 of the APC gene. This leads to an in-frame deletion, preserving the integrity of the reading frame. This variant has not been reported in the literature in individuals with a APC-related disease. This variant removes a portion of the armadillo-repeat region of APC (PMID: 11257105), which is suggested to be involved in the regulation of the actin cytoskeleton and cell-cell adhesion (PMID: 14672538, 17881494). The impact of a deletion within this region on APC tumor suppressive function is unknown (PMID: 11257105). In summary, this is a novel in-frame deletion with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance.
