Pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.10:g.(?_23626236)_(23630469_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 5-7 of the PALB2 gene. This creates a premature translational stop signal and is expected to result in an absent or disrupted protein product. While this particular deletion has not been reported in the literature, truncating variants in PALB2 are known to be pathogenic (PMID: 25099575, 17200668). For these reasons, this variant has been classified as Pathogenic.
