Pathogenic for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000011.10:g.(?_47331845)_(47336011_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 26-34 of the MYBPC3 gene. The 5' boundary is likely confined to intron 25. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. This variant has not been reported in the literature in an individual with a MYBPC3-related disease. This deletion eliminates the last 408 amino acids of the MYBPC3 protein (p.Gly868-p.Gln1274), which encompasses important functional domains of the protein and includes the region where several pathogenic missense variants have been described (PMID: 24793961, 25637381, 26358504). For these reasons, this variant has been classified as Pathogenic.
