NM_003070.5(SMARCA2):c.2612A>G (p.Tyr871Cys) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.2612A>G (p.Y871C) alteration is located in exon 18 (coding exon 17) of the SMARCA2 gene. This alteration results from an A to G substitution at nucleotide position 2612, causing the tyrosine (Y) at amino acid position 871 to be replaced by a cysteine (C). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
