Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005902.4(SMAD3):c.1032C>A (p.Asn344Lys), citing Ambry Variant Classification Scheme 2023: The p.N344K variant (also known as c.1032C>A), located in coding exon 8 of the SMAD3 gene, results from a C to A substitution at nucleotide position 1032. The asparagine at codon 344 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
