Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.8144T>A (p.Leu2715His), citing Ambry Variant Classification Scheme 2023: The p.L2715H variant (also known as c.8144T>A), located in coding exon 54 of the ATM gene, results from a T to A substitution at nucleotide position 8144. The leucine at codon 2715 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
