Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.8000T>G (p.Met2667Arg), citing Ambry Variant Classification Scheme 2023: The p.M2667R variant (also known as c.8000T>G), located in coding exon 53 of the ATM gene, results from a T to G substitution at nucleotide position 8000. The methionine at codon 2667 is replaced by arginine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
