Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.6e-05 in 251238 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. The c.346-10C>T variant has been observed in the presumed heterozygous state in at least 1 individual affected with Hereditary Breast And Ovarian Cancer Syndrome (example, Loveday_2011), without strong evidence for causality. To our knowledge, no experimental evidence demonstrating its impact on protein function has been reported.
