Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004168.4(SDHA):c.1695_1702del (p.Leu566fs), citing Ambry Variant Classification Scheme 2023: The c.1695_1702delCCTGGAGC pathogenic mutation, located in coding exon 13 of the SDHA gene, results from a deletion of 8 nucleotides at nucleotide positions 1695 to 1702, causing a translational frameshift with a predicted alternate stop codon (p.L566Afs*34). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
