Likely pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004168.4(SDHA):c.202G>C (p.Gly68Arg), citing Ambry Variant Classification Scheme 2023: The p.G68R variant (also known as c.202G>C), located in coding exon 3 of the SDHA gene, results from a G to C substitution at nucleotide position 202. The glycine at codon 68 is replaced by arginine, an amino acid with dissimilar properties. This variant was reported in individual(s) with features consistent with SDHA-related hereditary pheochromocytoma-paraganglioma (Ambry internal data). Based on internal structural analysis, this variant is anticipated to result in a significant decrease in structural stability (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
