Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_152703.5(SAMD9L):c.656A>T (p.Asn219Ile), citing Ambry Variant Classification Scheme 2023: The p.N219I variant (also known as c.656A>T), located in coding exon 1 of the SAMD9L gene, results from an A to T substitution at nucleotide position 656. The asparagine at codon 219 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
