NM_001754.5(RUNX1):c.281G>A (p.Ser94Asn) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S94N variant (also known as c.281G>A), located in coding exon 3 of the RUNX1 gene, results from a G to A substitution at nucleotide position 281. The serine at codon 94 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
