Uncertain significance — the classification assigned by Ambry Genetics to NM_133510.4(RAD51B):c.1035A>T (p.Gln345His), citing Ambry Variant Classification Scheme 2023: The p.Q345H variant (also known as c.1035A>T), located in coding exon 9 of the RAD51B gene, results from an A to T substitution at nucleotide position 1035. The glutamine at codon 345 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
