NM_000314.8(PTEN):c.190C>G (p.His64Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H64D variant (also known as c.190C>G), located in coding exon 3 of the PTEN gene, results from a C to G substitution at nucleotide position 190. The histidine at codon 64 is replaced by aspartic acid, an amino acid with similar properties. Other variant(s) at the same codon, p.H64P (c.191A>C), have been identified in individual(s) with features consistent with PTEN hamartoma tumor syndrome (Ambry internal data). In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
