Submitter rationale: The p.E235D variant (also known as c.705A>C), located in coding exon 7 of the PTEN gene, results from an A to C substitution at nucleotide position 705. The glutamic acid at codon 235 is replaced by aspartic acid, an amino acid with highly similar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally deficient (Mighell TL et al.
