NM_000264.5(PTCH1):c.2710A>T (p.Lys904Ter) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K904* pathogenic mutation (also known as c.2710A>T), located in coding exon 17 of the PTCH1 gene, results from an A to T substitution at nucleotide position 2710. This changes the amino acid from a lysine to a stop codon within coding exon 17. This variant was reported in individual(s) with features consistent with PTCH1-related nevoid basal cell carcinoma syndrome (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
