Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.3211G>A (p.Asp1071Asn), citing Ambry Variant Classification Scheme 2023: The p.D1071N variant (also known as c.3211G>A), located in coding exon 26 of the POLE gene, results from a G to A substitution at nucleotide position 3211. The aspartic acid at codon 1071 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
