Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.1294G>A (p.Glu432Lys), citing Ambry Variant Classification Scheme 2023: The p.E432K variant (also known as c.1294G>A), located in coding exon 12 of the PLOD1 gene, results from a G to A substitution at nucleotide position 1294. The glutamic acid at codon 432 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
