NM_001166108.2(PALLD):c.3316G>A (p.Glu1106Lys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E602K variant (also known as c.1804G>A), located in coding exon 10 of the PALLD gene, results from a G to A substitution at nucleotide position 1804. The glutamic acid at codon 602 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
