NM_001166108.2(PALLD):c.1965-12585C>G was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S149W variant (also known as c.446C>G), located in coding exon 1 of the PALLD gene, results from a C to G substitution at nucleotide position 446. The serine at codon 149 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
