Likely benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001035.3(RYR2):c.1022G>C (p.Gly341Ala), citing LabCorp Variant Classification Summary - May 2015: Variant summary: RYR2 c.1022G>C (p.Gly341Ala) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 0.00041 in 248946 control chromosomes, predominantly at a frequency of 0.0029 within the Latino subpopulation in the gnomAD database. The observed variant frequency within Latino control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in RYR2. c.1022G>C has been reported in the literature, without strong evidence for causality (Landstrom_2018). These report(s) do not provide unequivocal conclusions about association of the variant with Arrhythmia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
