This variant lies in the ACTN2 gene (transcript NM_001103.4) at coding-DNA position 892, where C is replaced by T; at the protein level this means replaces arginine at residue 298 with cysteine — a missense variant. Submitter rationale: Variant summary: ACTN2 c.892C>T (p.Arg298Cys) results in a non-conservative amino acid change located in the Spectrin repeat (IPR002017) domain of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 246158 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.892C>T in individuals affected with Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported.
