This variant lies in the ODAD2 gene (transcript NM_018076.5) at coding-DNA position 1850, where G is replaced by T; at the protein level this means replaces serine at residue 617 with isoleucine — a missense variant. Submitter rationale: This sequence change replaces serine with isoleucine at codon 617 of the ARMC4 protein (p.Ser617Ile). The serine residue is moderately conserved and there is a large physicochemical difference between serine and isoleucine. This variant is present in population databases (rs754364124, ExAC 0.01%) but has not been reported in the literature in individuals with a ARMC4-related disease.
