Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.746T>C (p.Phe249Ser), citing Ambry Variant Classification Scheme 2023: The p.F249S variant (also known as c.746T>C), located in coding exon 5 of the NOTCH1 gene, results from a T to C substitution at nucleotide position 746. The phenylalanine at codon 249 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
