Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.1275A>C (p.Glu425Asp), citing Ambry Variant Classification Scheme 2023: The p.E425D variant (also known as c.1275A>C), located in coding exon 9 of the APC gene, results from an A to C substitution at nucleotide position 1275. The glutamic acid at codon 425 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this variant remains unclear.
