Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4221G>T (p.Met1407Ile), citing Ambry Variant Classification Scheme 2023: The p.M1407I variant (also known as c.4221G>T), located in coding exon 22 of the DICER1 gene, results from a G to T substitution at nucleotide position 4221. The methionine at codon 1407 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
