Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "deleterious", MutationTaster: "polymorphism", Align-GVGD: "Class C65"). This variant is present in population databases (rs755140795, ExAC 0.005%) but has not been reported in the literature in individuals with a AKAP9-related disease. This sequence change replaces serine with cystine at codon 144 of the AKAP9 protein (p.Ser144Cys). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and cystine.
