Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.5513C>T (p.Ser1838Phe), citing Ambry Variant Classification Scheme 2023: The p.S1817F variant (also known as c.5450C>T), located in coding exon 37 of the NF1 gene, results from a C to T substitution at nucleotide position 5450. The serine at codon 1817 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
