Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.425G>C (p.Gly142Ala), citing Ambry Variant Classification Scheme 2023: The p.G142A variant (also known as c.425G>C), located in coding exon 3 of the FBN2 gene, results from a G to C substitution at nucleotide position 425. The glycine at codon 142 is replaced by alanine, an amino acid with similar properties, and is located in the EGF-like #02 domain. This amino acid position is well conserved in available vertebrate species; however, alanine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
